The people you send mail to are not going to have that file on their computer. If you want an image in your mail signature you're going to have to either attach it to every email (please don't) or have it hosted somewhere (which most email clients will block by default anyway). Alternatively just don't have an image in your email signature.

I found out that the solution is undoing the Add new buttons to Mail's preference panels hint. This hint showed how we could add a set of Apply/Cancel/OK buttons to Mail.app dialog windows. It looked innocent enough, but broke the signatures as described above. I don't know whether this only happened with the latest upgrade or before. May 14, 2018 Open the Mail app in Mac OS. Pull down the “Mail” menu and go to “Preferences”. Click the “Viewing” tab. Uncheck the box for “Load remote content in messages”. Exit out of Mail Preferences. Going forward, all new inbound email messages with any remote content, images, read receipts and email open confirmations, rich email HTML signatures with images, and other email tracking and remotely. I had the problem that Mail.app would no longer save any signatures. I could create them and use them as long as Mail was running, but they disappeared I soon as I quit Mail. I found out that the solution is undoing the Add new buttons to Mail's preference panels hint. To change these preferences in the Mail app on your Mac, choose Mail Preferences, then click Signatures. If you use iCloud Drive, your signatures are available on your other Mac computers that have iCloud Drive turned on. See Use iCloud Drive to store documents.

Wow.. thanks! This one has been killing me for weeks. I had resorted to creating .sigs on other machines and bringing the file over to mine.
There just *might* be some other problem (and solution :-) at hand here. Some weeks ago I used a homemade AppleScript to create some signatures. The signatures weren't being saved, although they were created. I have *never* applied the buttons-hack. The AppleScript works 100% OK and I have been using it for quite a while. My 'solution' was to manually, visually, check that signatures were created. Only then would they be saved.
I did apply the button hack and this signature bug did affect me.

I have not installed this hack and yet i have this identical problem..i cannot save Sigs after quitting Mail..any other ideas?
Yikes. Today I fixed it the hard way. During and after a one hour call to Apple technical support I did an Archive and Restore of the OS. That fixed the problem. PITA..
In my case, I did deploy the button hack, but the OS archive and install should fix the issue for those who did not deploy it also, since it replaces your busted Mail.app with a new copy.
Have you received a fix for this? I am experiencing the same loss of signatures created in mail 1.3.9. I also did not apply the button hack. If you know of a fix please let me know

1. Adding Signature to Email Accounts
Follow the steps outlined below to setup email signature on your Mac.
2. Using Multiple Email Signature
Mail app allows you to have multiple signature for single email account. You can create different signatures under “On My Mac” section and drag and drop the signature to required email account in the list. Alternatively, you can directly create new signatures for individual email account by selecting the email and click on + icon.
3. Using Image Signature
The email client on Mac is more flexible than the iPhone’s. As a result, not only you can add multiple text signatures, but you can also use images. To do this, just drag and drop the image into the signature box. But don’t forget to resize the picture before adding to the box. You can also do markup inside the signature box.
4. Choosing Email Signature in Preferences
On the Mail preferences section, you can setup a default signature for each email account. You have the following options when clicking on “Choose Signature” dropdown:

You can also choose the signature when you create a new email or when you are replying and forwarding. Go back to you Mail app and click on “Compose new message” icon. Alternatively, go to “File > New Message” menu or press “Command + N” to open a new message.
Here you can choose the signature from the dropdown and set the image size of the signature if you use image file in the signature.
6. Other Signature Options
Mac allows you to keep the font type of the signature as same as your email font. Also ensure to check “Place signature above quoted text” to add the signature above the previous text when replying or forwarding emails.
Mail app will automatically add signature when you are creating a new email, replying or forwarding a received email.
7. Signature for iCloud
When you are using iCloud Drive, select “iCloud” option and setup email signature as explained above. This signature will be available on other Mac devices when you use the same iCloud account.
8. How to Use HTML Signature?
Well, text and images are long gone. Now you need to add social icons and other HTML stuffs on the signature to identify yourself. There are no options in Mail app to insert HTML template signatures.
You need use third party apps for this purpose. Go to App Store on your Mac and search for “html mail signature” apps. You will find few premium apps depending upon your App Store country. Purchase an app and create templates as you need. Remember, you need to design HTML template on your own. The app will only allow you to add signature file in the Mail app library. The design and look of the signature is completely depends on your HTML code.
